چکیده

A simple and sensitive method for the analysis of oxytocin was developed using automated on-line in-tube solid-phase microextraction (SPME) coupled with liquid chromatography-tandem mass spectrometry (LC–MS/MS). Oxytocin was separated within 3 min on a Zorbax Eclipse XDB-C8 column, with water/methanol (10/90, v/v) as the mobile phase at a flow rate of 0.2 mL min−1. Electrospray ionization conditions in the positive ion mode were optimized for MS/MS detection by multiple reaction monitoring. The optimum in-tube SPME conditions were 20 draw/eject cycles of 40 μL sample at a flow rate of 250 μL min−1 using a Supel-Q PLOT capillary column as an extraction device. The extracted oxytocin was easily desorbed from the capillary by passage of the mobile phase, and no carryover was observed. The calibration curves for oxytocin were linear (r = 0.9981) in the range of 0−5.0 ng mL−1, and the relative standard deviations at each point were below 14.7% (n = 3). The limit of detection of this method was 4.0 pg mL−1, and its sensitivity was 58-fold higher than that of the direct injection method. This method was applied successfully to the analysis of oxytocin in saliva samples without any other interference peaks.

ON-LINE SOLID-PHASE EXTRACTION AND LIQUID CHROMATOGRAPHY/PARTICLE BEAM-MASS SPECTROMETRY FOR DEGRADATION STUDIES OF SOME POLAR PESTICIDES IN WATER

An on-line automated method for photodegradation studies of isoproturon, diuron, atrazine, fenitrothion, and metoxuron by means of liquid chromatography/mass spectrometry (LC/MS) with particle beam (PB) interface is described. Surface water samples were first spiked with 50 µg/l of each pesticide and then exposed to the radiation of the medium-pressure mercury lamp. Next, in regular intervals o...

Simultaneous Analysis of Metabolite Residues from Degradation of Dithiocarbamates in Cereals Using Liquid Chromatography Coupled with Tandem Mass Spectrometry

Background and Objectives: Dithiocarbamates are groups of fungicides used to control pests of agricultural products. Analysis of these pesticides with liquid and gas chromatography instruments is associated with various problems due to the presence of metal groups in the structure of these compounds. Therefore, one of the ways for analysis the pesticides is to check the level of decomposition...

Microextraction techniques coupled to liquid chromatography with mass spectrometry for the determination of organic micropollutants in environmental water samples.

Until recently, sample preparation was carried out using traditional techniques, such as liquid-liquid extraction (LLE), that use large volumes of organic solvents. Solid-phase extraction (SPE) uses much less solvent than LLE, although the volume can still be significant. These preparation methods are expensive, time-consuming and environmentally unfriendly. Recently, a great effort has been ma...

Determination of Two Antiepileptic Drugs in Urine by Homogenous Liquid-Liquid Extraction Performed in A Narrow Tube Combined With Dispersive Liquid-liquid Microextraction Followed by Gas Chromatography-flame Ionization Detection

A simple and efficient homogenous liquid-liquid extraction method performed in a narrow tube combined with dispersive liquid-liquid microextraction method has been presented for the simultaneous determination of two antiepileptic drugs in urine followed by gas chromatography with flame ionization detection. In this method, a mixture of acetonitrile and urine sample (homogenous solution) is load...
